'm interested in playing as a mage/assassin in Skyrim. What skills should I put the most perks in, and what dagger is the best blade? Is it the one you get in the Dark Brotherhood? How/where do I make/get a daedric dagger? And what enchantments do you recommend me getting on this dagger? Should I wear Archmages robes, or enchant my own armor? I'm also tired of my magika going down. I can only cast one or two spells before it all gets drained out. :/

There is a Daedric artifact weapon called Mehrunes' Razor that can instant kill.
Being a mage is fun, but not very effective. The strongest, most effective character to be in Skyrim is a Warrior/Paladin type. I have very high magicka, enchanted armor, and master level destruction spells, but these are still not very effective when trying to defeat the toughest opponents, like legendary dragons or dragon priests. Unless you want to be hard on yourself and try to only be a mage, the smithing skill is one of the most important skills to have.
For Smithing
Go to Whiterun and to the right there is that forge with the lady and her husband inside. Buy all the leather and leather straps you can from both the wife and husband and then smith as many leather bracers as you can. Then sell these back to her or drop them or wear them- nobody cares, but keep smithing these until your smithing is at 100. Then you can make dragon weapons. *you must have the Dawnguard DLC for Dragon Weapons* (if you don't have the DLC, Daedric weapons are the strongest)
Making the Dagger
For a dragon bone dagger, you will need one dragon bone, one ebony ingot, and leather strips. For the daedric dagger, you need an ebony ingot, a daedric heart**, and possibly some leather strips? After you make you dagger, take it to the grind wheel that's at this same forge and improve it with one dragon bone for the dragon dagger or one ebony ingot for the daedric dagger. NOTE: To improve your dagger to the max, you will need to create a full set of armor that has the "Fortify Smithing" enchantment and then wear it while you improve it. Also, taking a Blacksmith's Philter or Fortify Smithing potion before you improve adds to the overall damage of your dagger.
** For a daedric heart, go into the College of Winterhold and turn left into the Hall of Attainment. then go upstairs and find an elf named Enthir. You can buy two from him every 48 hours. They are expensive, though.
Enchanting the Dagger
Use a black or grand soul gem and the fire damage enchantment for max damage. This will not do an extremely large amount of extra damage, but it helps.
For the Assasin Role
You will need a high sneak skill, which can be improved my doing stuff while crouching or sneaking or whatever. You also will need the lock picking skill and the pickpocketing skill, which can be improved by doing these two things. I highly recommend joining the thieves guild in Riften, as their missions will give you places to practice these skills. They also give you armor like the Nightingale Boots that make your footsteps silent if you complete enough of their questline. Also, the one handed skill is very valuble, since the dagger you plan to use is a one handed weapon. This improves when you kill anything with a one handed weapon. Baisicly the entire game provides opportunities to level that up.
For Mage Role
Go to the College of Winterhold for all the spell tomes and such you will need. For master level spells, you have to do quests that are given to you by the experts in that magic category. For example, Frolda gives you the quest for the destruction magic's master spells once you learn all the other destruction spells. I use enchanted armor when i do magic, such as regen boosting amulets or enchanted armor that fortifies magic. However, the most helpful thing to do is take one piece of armor and enchant it with a magicka regen enchantment and maybe another and enchant it to give you more magicka, but the rest of your armor set should be enchanted with the "Fortify (Magic Type)" enchantment. that way the magic from that school will cost you very little. I used the fortify destruction enchantment on mine and I can kill giants and drauger deathlords without having to recharge my magicka bar. I also do not get killed easily, even by legendary dragons, since it is heavy armor and not a mage robe. The dark brotherhood quests may also be helpful for stealthy armor or something else that you might want. And when you do those quests you'll have enough gold to never worry about money again.
