i agree with the first person.... but would also add checking the distributor cap and rotor, the plug connection between the cap and the plug wire, and then the plug wire to the spark plug..
try changing one of the wires out with another and see if the cylinder you change it to loses spark... if so, then you need new wires...
are they new plugs? you could test this same theory with the plugs. put that plug in another cylinder and see if it misfires..
by the way, you need to take that plug out and clean it anyway. if it isn't firing, chances are its fowled out and may never fire unless you clean it.
if your distributer cap is is bad, it'll need to be replaced. same with the rotor.
I don't know what you have this motor in, but i would highly suggest that your first upgrade is an electronic ignition from mallory.
If you're not familiar, basically, there's a box which controls everything, sends it what is in essence an electronic distributor which takes place of your current one, sends spark to the plugs via wires.. nothing TOO fancy.
This will not only improve gas mileage, but puts a hotter spark through your plugs, thus burning the fuel more completely generating more power, and presumably less smog. it typically makes it a hair easier to start and stay started too - assuming your carb is adjusted right.
it's a very worthwhile investment...
if you can't figure this problem out, that may be your best bet.... to gut it and stick one of these on it.
I agree with everyone above. but if none of that works try adjusting tha valves thats what i had to do with my 350. just not to tight or you will bend tha rod and have to replace it
going along with the guy above me i would also do a compression check if the compression is low or bad the rings could be bad or there is a vaccume leak

well see it you are getting spark,if not could be bad plug, wire,or dirty points,
