Both these programs take quite a while to run, and this can be very boring and time-consuming. I was wondering if it was possible to run these two programs at the same time, or would they clash?

disk De-fragmentation means rearranging clusters of same file in order ( at a same sector on Your HDD) so that they can be accessed faster , it is a low level task and takes more time
Disk clean up means deleting temporary files created ...
And more a lock is applied on partition (drive) on which you r doing disk De-fragmentation so you can't access that partition while doing disk De-fragmentation
so it is not possible to do disk clean up disk De-fragmentation at a time
you can use third party tools to do automated disk De-fragmentation
you can use cleanup for fast clean up
Use Diskeeper after the disk cleanup, and you can use your computer as usual when it's running.
First do a disk cleanup. Right click the drive in My Computer, choose Properties and Disk cleanup. Delete the temp files, temporary internet files, empty the recycle bin etc. Don't compress the drive contents. It will take only a few minutes. Don't use the computer while it's running.
Head over to http://www.diskeeper.com/ and download the free trial version of Diskeeper 2010. It's the best defragger for Windows and defrags everything including large files and system files, even with low free space. Most importantly, it prevents up to 85% of fragmentation from occurring in the first place.
Install Diskeeper, turn on the auto defrag mode; Diskeeper will do everything in the background automatically, and you can use your computer as usual.
You should perform disk cleanup first and then disk defragmenter.
As a matter of fact, these programs provided by Microsoft are rather slow. If you want to save your time and energy, you should turn to other third-party tools.

The answer to this would be open to some debate. IMO, its a matter of preference, though I would strongly recommend that if you have been manipulating a large amount of files on your computer (creating, deleting, moving), and if its been more than a couple weeks since your last cleanup/defrag, it certainly wouldn't hurt to run it. You can set these operations as a scheduled task and schedule as often as you see fit...but at least once every week or two should be fine. :)

Run diskcleanup first and then defrag. You wouldnt want to waste time defragging files that need to be deleted anyway. Plus if you want the easiest and most effortless way to defrag, get Diskeeper2010, its the most advanced defragger which prevents most fragmentation from occuring. It can be set to run almost invisibly in the background and tackles any little fragmentation occuring in real time. Also defrags with less than 1% free space.
